In general, no not easily. SQLite lacks some features of MySQL that, while not essential for most database usage, are used in a lot of CMS systems. Also, the oft-used auto-increment feature has a different syntax for SQLite.

You'll almost certainly have to edit some code. If the CMS does it properly and uses a database abstraction layer like PDO or their own layer (or both), you won't have to go about searching for every single call of mysql_query (a function that should never really be used anywhere now that PDO exists).
